Narishige's NMN-21 is a manually operated micromanipulator which minimizes jitter for simplicity and high performance.
Drift and electromagnetic disturbance are critical problems in patch clamping work. This mechanical micromanipulator was developed specifically for patch clamping: it does not employ a liquid hydraulic system (which is prone to drift due to temperature changes), has no springs (a common cause of mechanical drift), nor a motor (which can be a source of electrical noise). Its body is designed to absorb vibrations, so that even at high magnifications, problems of jittering are virtually non-existant. The NMN-21 is based on a drum type design and utilizes a pair of independent sliders that serve to isolate vibrations. In addition, vibrations are often caused by the operator handling the control knob, which is usually connected to a slider. To overcome this problem, the control knob of the NMN-21 is fixed independently and separate from the slider. High mass and a low center of gravity provide exceptionally steady and stable performance. The overall design is compact, so installation requires a minimum of space. With coarse and fine manipulators integrated into the compact body, this model handles a full range of required motions, from moving a microelectrode into the microscope field of view to making delicate contact with cells.

The MM3A is also ideal for patch clamp experiments in the neurobiology lab. Below is a diagram of how the controls of the MM3A work. The NanoController and the Joypad (top two devices shown in the diagram) are included with the MM3A. Under the standard factory settings, the buttons on the joypad are used to move in coarse mode. The two joysticks can control in either fine or coarse mode depending on your speed selection. Additionally, you can set a home / park position that the micromanipulator can move quickly to and from when performing multiple injections.
For easier high-precision control (such as positioning a patch clamp pipette), we recommend the Cube, a manual control device with three high-resolution vernier dials (bottom device shown on the diagram). It gives you highly responsive, intuitive, linear movement control. It connects directly to the NanoControl control electronics in addition to your choice of joystick.

The Lift-Out Shuttle is a super-fast, dedicated lift-out solution for preparing samples for ex-situ electron microscopy. It features a microgripper, a four-axis substage, and a small CCD camera at surface level that allows for fast approach.
The system is comprised of a four-axis substage mounted on an ultra-compact platform. A microgripper is mounted above the substage to facilitate the lift-out. The stage is manipulated to position the pre-cut sample under the microgripper. After that, the microgripper clamps onto the sample and holds it gently, but firmly enough that the sample can be freed from the bulk material by simply dropping the stage down and out of the way. Narishige's PC-100 glass micropipette puller is a dual-stage vertical pipette puller system that provides reliable and consistent production of micropipets / glass needles with tip sizes ranging from 10 microns (10µm) to tenths of a µm.
The PC-100 incorporates several key features that make it an ideal choice for producing microinjection needles and pipettes for patch clamp experiments. The built-in power source is specially stabilized to deliver a uniform amount of current to the heating coil each time to make uniform glass needles and pipettes. The pulling force is provided by the earth's gravity, so you can't get much more stable or reliable than that. There are no electromagnetic coils to burn out. The force can be varied by adding or removing the included weight blocks (all of which are shown installed in the photo).
Heater levels for both single and double pull operations can be adjusted, and the heater level is shown by a highly-visible digital display.
During the pulling process, micropipettes are shielded from air currents by the use of an acrylic shroud. This excludes external drafts that might otherwise affect the shape and size of the micropipette.
Two pairs of weights (two light and two heavy) are provided for adjustment of the pulling force. Also included are a spare heater coil (PC-10H), and a sample of 1.0mm I.D. glass microcapillary tubes with internal glass fiber (GD-1).
Narishige now offers the more automatic and full-featured PC-100 to replace the old PC-10, PP-830, PP-83, and even older model PB-7. It serves as a lower-cost and more robust alternative to Sutter's P-30. In fact, we use the PC-100 here at Tritech Research to pull our pre-pulled microcapillary needles (see MINJ-PP) that we sell for C. elegans, Drosophila, Xenopus, Zebrafish, and transgenic mouse work.

Q: How can I troubleshoot my broken PC-100 to find out what the issue is? For example, if the display only reads 0.0 and will not increase nor will the coil heat when the red button is pushed.
A: First, for doing your testing, we suggest using the "Heater" setting on the selector switch, rather than the Step setting, so that the heater will be on all the time and not be dependent on the button push, carriage position, etc. A blown fuse would result in no display at all. You can see what happens by removing the fuse and comparing to see if the fuse is currently "working".Checking / changing the heating coil is a great place to start. If it isn't making good contact on both sides or is broken or nearly broken, that would definitely cause a problem and it might be intermittent at first.
Q: After recently replacing the coil and adjusting the heaters, my PC-100 is still having issues. I am not able to pull any decent pipettes. What could be the issue?
A: In general, higher temperatures will give a longer, finer tip and lower temperatures will give a more blunt tip. Please also check that the new heater is positioned so that the glass tube goes exactly down the middle of the coil (for even heating).
Q: How can I set the parameters of the PC-100 to achieve a specific type of needle tip?
A: There are many variables to get a particular size needle opening equivalent to a particular resistance. These include not only the settings of the puller, but also the glass used, the type of heater used in the puller, its condition, etc. Our main suggestion is that you contact a lab that is doing similar experiments and try to replicate their protocol for preparing your pipettes. The PC-100 can do both single and double pulls, so I'd start by finding out with is better / more typical. Generally speaking, a higher heater setting will give a longer thinner tip, and a lower setting will give a shorter tip that goes from the full size to a point over a shorter distance. Using all of the weights makes the tip get fine and be sharp quickly, and using less weights makes the tip longer and more gradual. The tip may come out of the puller with the right size opening, or sometimes the tips will come out completely sealed, and you will have to perform a secondary operation to open the end of the pipette to the size desired. For example, you can use the acid etching protocol described in the FAQ section at the bottom of this page.
Q: Our PC-100 has stopped working and seems to be overheating. What are some troubleshooting tips?
A: The first thing we recommend is checking the heating element itself. Make sure that it is not bent or touching back on itself (if you have the coil type). Also make sure that it is still perfectly centered around the pipette, and not nicked.
Q: How can I replace the coil on the PC-100?
A: First make sure the unit is unplugged / off, because the heater is pre-heated even between pulls, it's just not orange-hot. There are little metal bars that hold the heater on each side. You loosen / remove them to change the heater. The tedious part is that when you put the new coil in, you have to carefully position it so that the capillary tube will go right through the center. This is done best by having a piece of glass in the unit before the final tightening of the bars.
Q: Can the PC-100 produce sharp electrodes needed for purposes with resistances around 50 MegaOhms?

Employment of the oil hydraulic system enables microelectrodes to be inserted by remote control, without vibration. The chamber method is used, so that slight movements by the subject animal will not dislodge the microelectrode, while attachment/ detachment can be done freely. The acrylic XY stage makes it easy to position the microelectrodes with great accuracy while visually confirming their location. Microelectrodes ranging from 0.3 to 2mm can be used, by adjusting the positions of the packing and the microelectrodes themselves. Since this model uses the microelectrode expose method, any kind of microelectrode can be used. When the chain bar is filled with Ringer's solution, brain activity stops and long time recording becomes possible.

Narishige's SM-25C thin micromanipulator which can place several microelectrodes close together. Narishige's A-1 Muscle Fixing Apparatus facilitates insertion of a microelectrode into muscle or nerve fibers by gripping both ends of the muscle or nerve fiber and holding it under tension by stretching both ends mildly.
Normally, insertion of a microelectrode into muscle or nerve fiber is very difficult. Narishige's A-1 apparatus makes it easy by gripping both ends of the muscle or nerve fiber, stretching them and then holding the specimen under tension. Using the tweezers mounted on the ball joint, both ends of fibers of any length can be held, and moved in the Y-or Z-axis. Also attached is a stand, which allows vertical adjustment and on which the specimen is placed.

Motor-drive Oil Hydraulic Fine Open-Type Micromanipulator System
MDO-974 is the motor-driven oil-hydraulic micromanipulator suitable for in-vivo experiments on middle-sized animals, mainly NHP primates. Based on the manual oil-hydraulic mechanism of MO-97, the hybrid model installed with stepping motor enables speed control as well as greater driving precision, which are impossible to be performed by manual model. As a merit of the hydraulic part that requires no electricity, the hydraulic-tubing line keeps some distance from the motor to the animal's head, so that the sound or noise from the motor is suppressed during the experiment. Operational setting for drive is easy to do, which is completed by entering speed directly, or entering distance and time. The range of drive speed is set as 0.1um/s to 500um/s. The speed can be changed on the way to the target, so you can make the electrode closer to the target with a faster speed, and then change it to slower speed as final small-step approach. The closed chamber is equipped with acrylic XY stage which control knobs drive 18mm on each axis.

Narishige's SM-11 is the most popular (and versatile) micromanipulator for stereotaxic instruments because it can be attached to any of our AP frame bars and provides multi-axis movement. A coarse movement unit with a long working range is attached to the Z-axis, in addition to the fine movement adjustments included for each individual axis. These fine movement units have scales with minimum increments of 10µm (i.e. the total of a single rotation, 500µm, divided into 50 gradations), so that precise adjustments of 10µm are possible. Narishige's SM-25A Outfitted Thin micromanipulator can place several microelectrodes close together. The SMM-100C is a new coarse manipulator for stereotaxic instruments focusing on driving outside the animal's brain. The driving by Z-axis control knob: approximately 4mm per rotation makes faster adjustments for leveling bregma and lambda, and faster retracting/returning from the dura mater, which will improve efficiency of the experiment. For operations inside the brain, One-axis Motorized Stereotaxic Micromanipulator MDS-1 and hydraulic manipulator can be added optionally. The X-axis is equipped with a coarse control knob, which is the same as the Z-axis. The Y-axis (anterior/ posterior direction) is equipped with a fine control knob. The upper body of SMM-100C is switchable in either AP or ML direction when using it with tilting angle.
* The minimum graduation of the X and Z-axis is 100µm. This manipulator is recommended to be used in combination with a fine micromanipulator (MDS-1, etc).

Set-up example with SMM-A
SMM-A is recommended for experiments: tilting in the ML direction. When the manipulator SMM-100C/200C is tilted in the ML direction, the end of the holder (electrode tip) may be far off from animal's head. The attachment SMM-A is designed to attach a holder on the side of manipulator, so it enables the holder position without operating the X-axis. To this end, the holder (electrode tip) can be set to the animal's head while maintaining driving distance of the X-axis. *The set-up example above: motorized MDS-1 and IMS-30 on SMM-100C installed with SMM-A.

Stereotaxic Workstation

Narishige's SN-1N is a versatile, popular stereotaxic instrument used for middle size animals. This stereotaxic instrument is for middle size animals such as cats, small dogs and monkeys. A mouthpiece, eye bar and ear bar are mounted on the U- frame, to enable the zero point of head -- its basic axis -- to be accurately fixed in position, which in turn facilitates fixing of the upper jaw, lower edges of the eye sockets and external ear canals.
A spinal cord fixer is also mounted. The AP frame bar, to which manipulators and the spinal cord fixer are attached, is 18.7mm square in shape, and sufficiently long for several accessories to be attached simultaneously. Loosening the rear fixing bracket in the back allows the U-frame itself to be moved, as well as the unit supported at three points. The instrument body is made of lightweight cast aluminum, and offers wide-ranging versatility, since many kinds of animals can be fixed simply by changing the adapters used for the three support points.

Narishige's SR-8N head holder is designed for chronic and repeated experiments. This is an upgraded version of the compact, stable stereotaxic instrument SR-6N developed for small animals, and can be used for chronic experiments.
Use of the guide pipe, which passes between the skull and the scalp, enables repeated fixations in the same position without using anesthesia. This allows both acute and chronic experiments to be performed without pain or damage to the animals.
